Lab 1 Assessment

Albumins are a type of water soluble protein primarily found in blood which also have a purpose in inducing the artificial capacitation of sperm. Capacitation is a process of the maturation of sperm occuring in the female reproductive tract in which the acrosomal head is distabilized to allow a greater degree of binding with sperm and oocyte. Other cellular changes such as the formation of ROS and protein phosphorylation of Tyrosine residues also occurs. This paper aims to measure the extent of capacitation of sperm via different serums through tests such as ROS Level Measurement, Sperm Mmotion Analysis, a Hemizona Assay, Acrosome Reaction Evaluation, Western Blot Analysis of Tyrosine Phosphorylation and Sperm DNA Damage Assessment. These values obtained via each test were then subjected to statistical analysis via Wilcoxon signed-rank test which determines whether population means differ between samples. 30 samples of semen were obtained from male specimens, each sample split up into 4 portions and subsequently cultured by 4 different preparation media; HTS, HTP, QFS and QTP respectively which used differing levels of SSS (Serum Substitute Supplement) and SPS (Quinns Advantage Serum Protein Substitute) which are both known spermatozoa capacitating agents. It was found that the Serum Supplements induced semen capacitation at varying levels, primarily influenced by ROS levels in preparation media and secondarily by the combination of Serum Supplements and Media used.

There is quite a bit of current research looking at what effects success rates in these clinical techniques. DMD is diagnosed through DNA tests searching for deletions, duplications and point mutations, a muscular biopsy is more conclusive than the DNA testing itself. In DMD, there is a lack of the protein dystrophin which leads to muscle deterioration and breakdown. An animal model used is the MDX mouse which has missing functional dystrophin protein in the muscle Dystrophin connects the cytoskeleton of the muscle fibre to extracellular matrix through the cell membrane.
