Introduction

There are many steps in the preparation and processing of tissue sections that can introduce artefacts into the original tissue. This page introduces some of the steps in tissue preparation where this can occur and the type of artefact that appears in the final tissue. The pages on histology fixatives and stains should be read in conjunction with this page.

Prefixation Artifacts

Anything that may have been done to the tissue before it has been preserved by fixation. This includes handling, physical distortion, drug treatments or injections, tissue degeneration, or poor tissue preparation for fixation.

Fixation Artifacts

Fixative solution osmolality

  • Hypertonic can cause cellular shrinkage and increased extracellular spaces.
  • Hypotonic can cause cell swelling.
