Mouse Embryo Lipid Droplets

Time-lapse of embryo development optical sections of the mouse embryo.

  • At fertilization, lipid droplets are small and uniformly distributed.
  • Implanting blastocyst lipid droplets are larger and enriched in the invading giant cells of the trophectoderm.
  • Movie starts at the compacted morula stage and development is followed till the blastocyst stage.
  • The majority of third harmonic generation (THG) signal is generated by lipid droplets, which become fewer and larger over the course of development.
  • Imaged with third harmonic generation (THG, grey) and second harmonic generation (SHG magenta).
  • Images were captured at 20 minute intervals. Scale = 20 μm.


Harmonic Generation Microscopy (HGM)

  • laser scanning microscope three-dimensional, label-free imaging of developing embryos
  • takes advantage of second and third harmonic generation (SHG and THG respectively)
  • result in the generation of photons of half and one-third the illumination wavelength respectively.
