Mouse Embryo Lipid Droplets

Mouse lipid droplets icon.jpg

Time-lapse of embryo development optical sections of the mouse embryo. At fertilization, lipid droplets are small and uniformly distributed, but in the implanting blastocyst, lipid droplets are larger and enriched in the invading giant cells of the trophectoderm.

  • Movie starts at the compacted morula stage and development is followed till the blastocyst stage.
  • The majority of third harmonic generation (THG) signal is generated by lipid droplets, which become fewer and larger over the course of development.
  • Imaged with third harmonic generation (THG, grey) and second harmonic generation (SHG magenta).
  • Images were captured at 20 minute intervals. Scale = 20 μm.

Harmonic Generation Microscopy (HGM)

  • laser scanning microscope three-dimensional, label-free imaging of developing embryos
  • takes advantage of second and third harmonic generation (SHG and THG respectively)
  • result in the generation of photons of half and one-third the illumination wavelength respectively.
