Wnt ligands which are able to activate the calcium ion pathway firstly physically interact with the Fz receptor on the surface of cell membrane. The signalling complex formed by Dsh and beta and gamma G-protein subunits then activates the PLC protein, which cleaves the membraneous protein PIP2 into IP3 and DAG. IP3 then activates the corresponding receptor on the endoplasmic reticulum leading to influx of calcium ions into the cell cytoplasm. Calcium ions then activate three important downstream targets, CaCN, CaMKII and PKC. through interaction with other proteins, calmodulin or DAG. A series of cellular events will then take place as a result of the activation of those mediators. Calcium ion pathway mainly regulates the binding of TCF/beta-catenin with the DNA to decrease beta-catenin-mediated DNA transcription through CaMKII. Moreover, CaMKII may induce gene transcription itself as well. CaCN mainly functions as a transcription factor, and PKC may play an important role in cell migration.
