Thalidomide destabilizes CD147 to cause Teratogenicity

a) Top, representative images of zebrafish larvae that were treated with either DMSO or 400 μM thalidomide (Thal) and analyzed at 3 d post fertilization (d.p.f.); anterior to the left, dorsal view. The dashed lines demarcate the eyes.

Thalidomide (and the derivatives lenalidomide and pomalidomide) are treatments for hematologic malignancies, multiple myeloma (MM) and del(5q) myelodysplastic syndrome (MDS). Cereblon (CRBN) is a substrate receptor of the CRL4 ubiquitin ligase complex and is the primary target by which they mediate anticancer and teratogenic effects. The ubiquitin-independent physiological chaperone-like function of CRBN that promotes maturation of the CD147 and solute carrier family 16 member 1 (SLC16A1; also known as MCT1) proteins. CD147

(basigin, BSG) Basigin is a member of the immunoglobulin (Ig) superfamily, with a structure related to the putative primordial form of the family.
